下記の通り、Michael Bassik, Ph.D.( Assistant Professor, Stanford University )のセミナーを二部構成で行なうことになりました。
Bassik先生は、初めて”systematic genetic interaction maps in human cells”の確立に成功するなど世界的な研究成果をこれまで数多く発信されてきています。最近では、”a double sgRNA platform”の開発に成功していて国際的にも高く評価されています。

演 題 | Parallel shRNA and CRISPR/Cas9 Screens Reveal Biology of Stress Pathways and Identify Novel Drug Targets. |

要 旨 | We have developed high-complexity shRNA libraries (25 shRNAs/gene) that greatly reduce false negatives/false positives, and have adapted these libraries to knock down gene pairs to perform systematic genetic interaction maps in mammalian cells. We have used these maps to study ER-trafficking toxins, and identified novel protein complexes as well as insights into retrograde trafficking. We are now using this strategy together with the CRISPR/Cas9 system to study stress signaling more globally, as well as to identify novel drug targets. In recent work, we have used parallel shRNA and CRISPR/Cas9 screening to explore the cellular response to oxidative stress, and have identified the target and mechanism of action of a novel host-targeting antiviral drug. |
