要旨
B cells play central roles in humoral immune responses. In the context of autoimmune disorders, B cells are widely accepted as pathogenic effectors through their production of autoantibodies. However, evidence is accumulating that B cells can also be immunosuppressive in autoimmune inflammatory diseases.
This subset of B cells with regulatory property, known as ‘regulatory B cells’, contributes to the inhibition of pathogenic immune reactions, primarily via the production of an anti-inflammatory cytokine, interleukin-10 (IL-10).
Although subpopulations of splenic B cells have been reported to express IL-10 in vitro, little is known about the identity of IL-10-producing regulatory B cells in vivo. We have recently shown that plasmablasts in the draining lymph nodes (dLNs) predominantly express IL-10 in vivo during experimental autoimmune encephalomyelitis (EAE).
These plasmablasts were generated only during EAE inflammation, which suppressed EAE development. Furthermore, human plasmablasts also preferentially secrete IL-10, further highlighting plasmablasts as the IL-10-producing regulatory B cells.
We will discuss how B cells produce IL-10 and negatively regulate pathogenic immune reactions.
