要旨

Faithful copying and maintenance of mitochondrial DNA (mtDNA) are fundamental to the oxidative phosphorylation and therefore to life. The importance of mtDNA maintenance can be recognised by the fact that defects in mtDNA are a common cause of human genetic disease. Also, possible links of ageing and mtDNA abnormality are implicated. For three decades a simple replication model for mammalian mtDNA had generally been accepted. A series of recent studies, however, seriously challenged the model and demonstrated that mitochondria employ complex DNA replication system with more than one replication mechanism, none of which is identical to the old model. The most striking feature of mtDNA replication is that significant proportion of the replication intermediates contain long stretch of RNA, which is unprecedented in DNA replication.
In the seminar I will briefly overview the property and the clinical relevance of human mtDNA and present our recent understanding of mtDNA replication.
